Product description

High-efficiency plasmid vector for cloning PCR products with T overhangs and blue/white screening capability.

Promega's pGEM®-T Vector System is a high-efficiency tool designed for the cloning of PCR products, particularly those generated by thermostable polymerases that add a single deoxyadenosine to the 3′-ends of amplified fragments. This vector system facilitates the rapid and reliable insertion of PCR products into plasmids, streamlining molecular cloning workflows.

Key features include:

  • Single 3′-T Overhangs: Enhance ligation efficiency by preventing vector recircularisation and providing compatible overhangs for PCR products with A overhangs.
  • Blue/White Screening: T7 and SP6 RNA polymerase promoters flank a multiple cloning region within the α-peptide coding region for β-galactosidase, allowing direct identification of recombinant clones through colour screening on indicator plates.
  • Rapid Ligation: The provided 2X Rapid Ligation Buffer enables ligation reactions to be completed in just one hour at room temperature, significantly reducing processing time.
  • Versatile Insert Release: The multiple cloning site is flanked by recognition sites for the restriction enzyme BstZI, allowing release of the insert by a single-enzyme digestion. Alternatively, a double digestion may be used to release the insert from the vector.
  • Single-Stranded DNA Preparation: The f1 origin of replication allows for the preparation of single-stranded DNA, facilitating applications such as mutagenesis and sequencing.

This vector system is widely utilised in various scientific fields, including:

  • Molecular Biology: For gene cloning and expression studies.
  • Genetic Engineering: In the development of recombinant DNA technologies.
  • Biotechnology: For the production of genetically modified organisms and proteins.

Commonly employed in:

  • University Laboratories: For academic research and teaching purposes.
  • Biomedical Research Centres: In the study of gene function and disease mechanisms.
  • Clinical Laboratories: For diagnostic and therapeutic research applications.

First introduced in the 1990s, the pGEM®-T Vector System remains a relevant and widely used tool in molecular cloning due to its efficiency and ease of use.

Why Questpair exists

Lab equipment markets are fragmented. Buyers spend weeks calling distributors, comparing quotes, and verifying supplier reliability. Sellers struggle to reach global research labs.

We built Questpair to fix this. Every supplier on the platform is verified before listing. Every quote you receive comes from a real, vetted seller. AI prepares your case in minutes, then a real specialist takes over.

We're an independent team based in The Netherlands, working with research labs and procurement teams across Europe and worldwide.

Frequently asked questions

How long does laboratory equipment typically last?

With a service contract, ten to fifteen years is normal for most benchtop instruments. Detectors and pumps are the parts that wear first.

Can I get service for older equipment?

Often, yes. Verified suppliers frequently work with service partners converting the instrument. If applicable, we can list servicing options alongside the offer.

Where can I buy parts?

Ask a scientist through the request button. Parts are often available even when the instrument itself is discontinued.

What is the typical lead time for refurbished equipment?

Two to six weeks, depending on refurbishment scope and whether calibration is included.

Does laboratory equipment ship internationally?

We ship to 38 countries. Crating and customs handling are quoted with the instrument.
