The Amersham Biosciences ÄKTAxpress is a liquid chromatography system specifically designed for automated, multi-step purification processes. It is engineered to efficiently purify Histidine-tagged and Glutathione S-Transferase (GST)-tagged recombinant proteins from either clarified or crude cell lysates. ÄKTAxpress accommodates purification protocols that include up to four chromatography steps such as affinity chromatography, ion exchange, desalting, and gel filtration.
It offers versatility with protocols tailored for both low and high expression proteins, as well as double-tagged proteins. The system also facilitates on-column and off-line (batch) tag cleavage processes. ÄKTAxpress can purify up to four samples concurrently, with each capable of processing up to 50 mg of protein.
Multiple ÄKTAxpress systems, up to twelve, can be managed in parallel from a single computer, allowing for simultaneous and varied protocol execution. The systems can be configured either adjoining or spaced apart according to laboratory preferences.
The system supports a flow rate range of 0.1 – 65 mL/min, with an accuracy of ±2% or ±0.02 ml/min, and a pressure range of 0 – 30 bar. It is capable of forming step gradients with water-based buffers and concentrations between 5 – 95%, ensuring precise and linear conditions.
The UV monitor operates at wavelengths of 280 and 254 nm, with an optical path length of 2 nm, ensuring absorbance within the range of 0.01 – 5 AU, and providing excellent measurement stability and accuracy.
The ÄKTAxpress features a conductivity range from 0.01 – 300.00 mS/cm, maintaining high reproducibility and minimal noise levels. The compact design measures 250 x 490 x 660 mm and weighs 30 kg, making it a robust addition to any laboratory focused on precise protein purification.
