The heart of the transducer is a lead zirconate titanate electrostrictive element that, when subjected to an alternating current, expands and contracts. The implosion of microscopic bubbles or voids in the solution results, intensively stirring the molecules in the medium. The Sonifier functions in two modes: Pulsed and Continuous. In Pulsed Mode, ultrasonic vibrations are transferred to a solution at a rate of one pulse per second. This pulse duration can be adjusted from 0.1 to 0.9 per second, allowing the solution to be processed at full ultrasonic intensity while the temperature builds up – particularly valuable when processing warm volatile solutions and temperature-sensitive material. Plating density can be as high as 500 colonies per 4×4 inch agar plate and typical colony diameters can be easily less than 0.5 millimeter. Blue/white selection and plaque picking are also available. The instrument is controlled with an intuitive graphical user interface, which provides complete real-time control of the intrument. The system has an on board imaging system for image acquisition. Higly optimized image analysis software automatically locates the position of objects of interest only. The image analysis software also allows individual selection, using a mouse, of any objext in the field of view for custom picking applications. Twenty tungsten needles, guided by precision linear slides, independently pick and place clones of interest. The input agar or microtiter plate is moved under the pick needle by high precision X,Y stages capable of positioning within 6 microns.
