The Nucleofector technology by Amaxa represents a significant breakthrough in the field of electroporation by combining optimized electrical parameters with specialized Nucleofector solutions tailored for various cell types. This advanced approach allows for the direct delivery of DNA into the cell nucleus, enabling immediate expression. As a result, nucleofected cells are ready for analysis after a brief incubation period of only 2 to 8 hours.

Ideal DNA quality parameters include a plasmid concentration of 1–5 µg per nuclefection sample (100 µl), which can be increased to 10 µg per sample. While this may result in higher gene transfer efficiencies, it could also lead to increased cellular mortality.

Recommended cell density for suspension cells ranges from 2 x 10⁵ to 1 x 10⁷ per nuclefection sample, while for adherent cells, it is between 2 x 10⁵ to 2 x 10⁶ per sample. It's advisable to avoid cell densities below 2 x 10⁵ cells per sample, as this may decrease gene transfer efficiency and increase mortality.
