The ÄKTAxpress is a highly efficient liquid chromatography system expressly developed for automating multi-step purification processes. It is adept at purifying Histidine-tagged and Glutathione S-Transferase (GST)-tagged recombinant proteins from both clarified and crude cell lysates. This intricately designed system can effectively handle purification protocols incorporating up to four distinctive chromatography steps such as affinity chromatography, desalting, ion exchange, and gel filtration, in that order. It successfully encompasses methods for purifying both low expressed and well-expressed proteins. Additionally, it enables purification of double-tagged proteins, and supports protocols that integrate with both on-column and off-line (batch) tag cleavage.
The key features of the ÄKTAxpress system include:
• Autonomy in purifying one to four samples concurrently in a single run.
• Can accommodate purification protocols with up to four chromatography steps.
• Automatic on-column affinity tag removal in the first step of protocols starting with an affinity/ion exchange chromatography.
• Supports purification of both well-expressed and low expressed proteins, as well as double-tagged proteins and proteins subject to off-line cleavage.
• Ability to purify up to 50mg protein per sample.
• Capable of running up to twelve ÄKTAxpress systems interactively, controlled from one computer (not included), with different purification protocols running simultaneously.
• Flexibility in positioning – systems can be located adjacently or placed separately as per preference.
Technical specifications:
• Flow rate: 0.1 – 65 mL/min with ±2% accuracy or ±0.02 ml/min, whichever is greater.
• Pressure range: 0 – 30 bar with an RSD of <0.5% for flow rate reproducibility.
• Allows step gradients and linear gradients conditions using water-based buffers with varying salt concentrations.
• Gradient mix proportions and concentration accuracy are 5 – 95% and ±2%B respectively.
• Affords UV monitor wavelengths at both 280 and 254 nm.
• Optical path length and cell volume are matched at 2 nm and 2 µL respectively.
• Absorbance range lies between 0.01 – 5 AU with auto zero range from -0.2 – 2.0 AU.
• Conductivity ranges from 0.01 – 300.00 mS/cm and the internal volume is 24 µl.
