The Büchi Reveleris X2 Flash Chromatography System is an advanced instrument designed for the swift purification of complex samples using liquid chromatography. It operates using a cartridge with a stationary phase like 40-63µm silica and a mobile phase mixture such as hexane: ethyl acetate (70:30), at flow rates up to 40mL/min and moderate pressures under 100 psig. Although the resolution is less compared to HPLC, where 2-5 µm particles are used, flash chromatography excels at separating gram-sized samples quickly.

The system's separations rely on the interaction between sample compounds, the stationary phase, and the mobile phase. Compounds retained longer on the stationary phase indicate stronger interaction compared to those with the mobile phase. Conditions for sample separation are typically determined through thin layer chromatography (TLC). Although normal phase cartridges are common, various other stationary phases are available.

The Reveleris X2 system integrates a UV detector capable of monitoring three wavelengths simultaneously, alongside an evaporative light scattering detector (ELSD), which can detect all non-volatile compounds in the mobile phase and compounds that do not absorb UV radiation. This allows for diverse detection even when mobile phases interfere with absorbance wavelengths of interest. Both detectors offer adjustable sensitivity for efficient detection and collection of both small and large compound quantities.

Key operational features include a gradient flash chromatographic system with a solvent delivery system comprising four independent channels and a binary gradient with high-pressure mixing. The system supports flow rates ranging from 1 to 200mL/min and delivers flow rate accuracy of ± 3% for flow rates between 4 and 200mL/min, with a maximum pressure of 200 psig. It boasts gradient accuracy of ± 1.5% and precision of < 1% at 40mL/min, with sample modes supporting both liquid and solid samples. The sample loader accommodates weights of 3g, 25g, and 60g, with plungers in three sizes (15mL, 75mL, and 150mL). The absorbance detector covers a wavelength range of 200 to 500 nm, monitoring up to three wavelengths concurrently to trigger fraction collection via UV or ELSD signals. Lastly, the multi-position collection racks are equipped to handle numerous tubes efficiently.
