The Büchi Reveleris X2 Flash Chromatography System is a sophisticated instrument designed for the efficient and rapid purification of complex samples via liquid chromatography. This system provides flash chromatographic separation with capabilities that include the use of cartridges containing stationary phases such as 40-63 µm silica coupled with mobile phases, like a hexane:ethyl acetate mixture (70:30), at flow rates of up to 40 mL/min and moderate pressures not exceeding 100 psig. While its resolution may not match that of analytical or preparative High-Performance Liquid Chromatography (HPLC), which utilizes smaller particles (2-5 µm), flash chromatography is invaluable for its ability to handle gram-sized samples swiftly and effectively.
As with analytical or preparative HPLC, flash chromatography relies on the differential interactions of sample compounds with the stationary and mobile phases. Compounds exhibiting stronger affinity to the stationary phase exhibit prolonged retention times compared to those favoring the mobile phase. Typically, preliminary conditions for separating the sample are determined through thin-layer chromatography (TLC). Although normal-phase cartridges are predominantly used, various other stationary phases are available to optimize separation.
The Reveleris X2 system enhances detection capabilities with a UV detector capable of monitoring three wavelengths concurrently, alongside an evaporative light scattering detector (ELSD), essential for detecting non-volatile compounds that are invisible under UV light. The ELSD permits the use of mobile phases that absorb light at wavelengths similar to the target compounds. Both detection systems feature adjustable sensitivity settings, enabling precise detection and collection of both minor and significant quantities of compounds.
