The Amersham Biosciences ÄKTAxpress FPLC System is an advanced liquid chromatography apparatus designed for automated, multi-step protein purification processes, making it highly valuable for research professionals handling complex biochemical analyses. This particular system excels in the purification of Histidine-tagged and Glutathione S-Transferase (GST)-tagged recombinant proteins derived from either clarified or crude cell lysates.


Key features of the ÄKTAxpress include the capability to execute purification protocols that incorporate up to four distinct chromatography steps. These can include affinity chromatography, desalting, ion exchange, and gel filtration, offering flexibility depending on experimental requirements. Within its protocols are options for handling both low and well-expressed proteins, as well as those requiring on-column or offline (batch) tag cleavage. Notably, the system supports the automated purification of up to four samples in a single run with an ability to remove affinity tags efficiently during the initial step when using an affinity/ion exchange approach.


The system is designed to purify up to 50 mg of protein per sample, and with a modular setup, up to twelve ÄKTAxpress units can be operated concurrently from a single computer (not included), allowing for simultaneous execution of diverse purification protocols. Its flexible configuration supports placement of units together or individually as per your laboratory's layout preferences.


Performance metrics include:

- Flow rate: 0.1 – 65 mL/min with an accuracy of ±2% or ±0.02 ml/min, whichever is greater, and flow rate reproducibility at RSD < 0.5%.

- Pressure range: 0 – 30 bar.

- Execution of step gradients with water-based buffers, providing gradient mix proportions of 5 – 95%, with concentration accuracy within ±2%B.

- UV detection at wavelengths of 280 and 254 nm, facilitated by 2 nm optical path length and 2 µL optical cell volume, featuring an absorbance range of 0.01 – 5 AU and linearity deviation < 3% to 2 AU.

- Conductivity readings ranging from 0.01 – 300.00 mS/cm with an accuracy of ±2% of full scale calibrated range or ±0.1 mS/cm, showing reproducibility and noise metrics that ensure precise readings during long or short-term experiments.

- The system dimensions are 250 x 490 x 660 mm with a weight of 30 kg, ensuring a compact footprint for modern laboratory environments.
