Roche LightCycler 96 Real-Time PCR System (Model 4393) is a benchtop thermal cycler designed for rapid PCR amplification and fluorescence detection with high sensitivity and precision.
- Fast PCR cycling: 40 cycles completed within 40 to 60 minutes.
- Control: Operates via touch panel; no PC required for running protocols.
- Data transfer: USB memory and LAN connection supported for data acquisition.
- Temperature control: Uses Peltier elements, with max heating at 4.4 °C/sec and cooling at 2.2 °C/sec; accurate to ±0.2 °C and uniformity ±0.3 °C over 37–98 °C.
- Gradient control: Available over a 20 °C range between 37 and 98 °C.
- Optics: High-power broad spectrum LEDs provide four excitation wavelengths (470, 533, 577, 645 nm) paired with four detection wavelengths (514, 572, 620, 697 nm); fixed excitation/detection combinations enable detection of dyes such as SYBR Green I, FAM, VIC, HEX, Texas Red, and Cy5 without ROX normalization.
- Multiplexing: Up to 4 colors per reaction.
- Detector: 96 × 2 channels using glass fiber optics and CCD camera for fluorescence detection.
- Sample throughput: 96 samples per run with compatibility for 0.1 mL block tubes, 8-strip tubes, and 96-well plates; reaction volumes from 10 to 50 μL.
- Noise level: Operating noise less than or equal to 43 dB.
- Software compatibility: Runs on Windows XP, 7, 8, 10, and 11 with comprehensive analysis options including absolute/relative quantification, melt curve analysis, genotyping, HRM, and qualitative detection.
- Physical dimensions: 400 mm wide, 400 mm deep, and 530 mm high; weight approx. 27 kg.
- Power: Requires 100 – 125 V AC, 600 W power supply at 50/60 Hz.
