The Roche LightCycler 96 is a dedicated real-time PCR system optimized for high-throughput, multiplex fluorescence detection essential in molecular diagnostics and biotechnology research. Featuring rapid thermal cycling with precise temperature control using a Peltier element, it achieves 40 PCR cycles in under 40–60 minutes, supporting workflows requiring fast turnaround. The system supports 96 samples per run using 0.1 mL blocks, 8-strip tubes, or 96-well plates with reaction volumes of 10–50 µL.

Equipped with an ultra-high brightness white LED excitation source and glass fiber optics coupled to a CCD camera, the instrument allows four-channel multiplexing with excitation wavelengths at 470, 533, 577, and 645 nm and emission detection at 514, 572, 620, and 697 nm. This supports common fluorescent dyes such as SYBR Green I, FAM, VIC, HEX, Texas Red, and Cy5, enabling flexible assay formats including intercalator and hydrolysis probe-based qPCR.
